Dr.Chufeng Li, received his M.S. degree in Condensed Matter Physics from Wuhan University in 2010 and Ph.D. degree in Biophysics from Arizona State University in 2016. From 2016 to 2019, postdoctoral Fellow in Arizona State University Biophysics Center/National Science Foundation (NSF) BioXFEL STC Program, focusing on Sequential femtosecond crystallography data analysis algorithm research and software development, and ultrafast kinetic two-color diffraction method development for biological macromolecules. 2019-present, Postdoctoral Research Associate at Free Electron Laser Science Center, Deutsches Elektronen-Synchrotron DESY (DESY), Germany, major in Coherent Convergent X-ray Crystallography, Diffraction Microscopy method development. His research has been published in IUCrJ, Nature and Nature Communications, et al.

摘要:

The superior brightness and short pulse duration of X-ray free electron laser (XFEL) allows it to outrun radiation damage in coherent diffractive imaging. In each imaging event, the elastic scattering terminates before photoelectron cascades commences. This“diffract-before-destroy” feature of XFEL opened new opportunities for biological macromolecule imaging and structure studies by breaking the limit to spatial resolution imposed by the maximum dose that is allowed. In this talk, we introduce typical XFEL diffractive imaging experimental schemes, and data collection modes including solution scattering, serial femtosecond crystallography (SFX), single particle diffraction, and mix-and-injection. We then go through specific aspects such as sample delivery and online data reduction and analysis pipeline. Finally, recently developed experimental methods and analysis algorithms are discussed which aim at increasing the data efficiency and advance the capability to fast dynamics studies.
